Abstract
To gain a better understanding of possible mechanisms by which glucosyl hesperidin(Ghesperidin)lowers serum triglyceride(TG)level, we carried out oral administration tests ofG-hesperidin in the rats fed a high-fat diet. Serum TG and(VLDL+LDL)-cholesterol levelsas well as hepatic TG and cholesteryl ester(CE)contents significantly decreased by Ghesperidinadministration. In the analysis of hepatic gene expression, G-hesperidin significantlydecreased mRNA levels of fatty acid synthase(FAS)and SREBP-1c;and moreover,this flavonoid caused the significant increase of carnitine palmitoyltransferaseⅠ, Ⅱ andPPAR-α mRNA levels. The same effects were also observed by hesperidin administration.These findings indicate that G-hesperidin behaves as hesperidin in the body and results indecreasing hepatic TG and CE through the down-regulation of FAS and the augmentation offatty acid β-oxidation. Furthermore, it is considered that this effect contributes to the suppressionof excess VLDL secretion in the liver, leading to the reduction of serum TG level.
