Abstract
Objective In this study, preventive effects of Agaricus blazei Murrill(Himematsutake)on oral mucositis(OM)induced by 5-Fluorouracil(5-FU)were examined. Methods Histopathological changes for OM were assessed in paraffin sections stained with hematoxylin and eosin(HE), according to the standard method. Hematological findings were estimated by fully automatic multi-item analyzer. Furthermore, differential leucocyte count (%)was examined with Giemsa staining under the microscope. In addition, histamine release from rat peritoneal mast cells was measured by fluorometric assay and antihistaminic action was examined by magnus modified method using small intestine of guinea pig. Results White blood cells(WBC)markedly decreased after 5-FU administration. OM were established in mice by intraperitoneal administration of 30 mg╱kg of 5-FU in three times every other day. OM were observed in 100% mice in this group. In combinational group(5-FU plus Agaricus blazei hot water extract(ABWE)), OM were observed in 20% mice and OM markedly decreased. The number of WBC decreased significantly by 5-FU. Large lymphocytes decreased in differential leukocyte count(%)by 5-FU. These suppression by 5-FU recovered to a normal level after oral administration of ABWE. ABWE inhibited histamine release from rat peritoneal mast cells and ABWE showed antihistaminic action in small intestine of guinea pig. Conclusions These results suggest that ABWE may have clinical value in the prevention of OM induced by 5-FU. (Jpn Pharmacol Ther 2022;50:1353‒60)
