Objectives Assess the anti−inflammatory and analgesic effect of Voltaren_tape(containing1% diclofenac sodium)and compare it with that of other NSAID−containing tapes. Methods The analgesic and anti−inflammatory effects of 1% diclofenac sodium(Voltaren_tape), 3.5% felbinac−containing tape, 0.5% felbinac−containing tape and 3.75% indomethacin−containing tape were evaluated using the carrageenan−induced hind footpad paw edemaand brewer’s yeast−induced hyper algesia models in rats. Tapes were applied at 2 hoursbefore injection of carrageen or brewer’s yeast. The volume of the right hind paw and painthreshold were assessed at 1, 3, 5 and 7 hours after induction of inflammation. Results The edema ratios at the Voltaren_tape group were lower than at the control group. The edema ratio at 5 hours after induction of inflammation and the AUEC(area under theeffect curve)were significantly lower than at the control animals(p=0.009). The inhibitoryratio in the Voltaren_tape group(12.1−33.2%)was higher than in the comparative groups. The pain threshold ratios measured at 3 and 5 hours after induction of inflammation were significantlyhigher and the 1/AUEC(reciprocal area under the effect curve)was significantlylower than the control group control(p=0.004). In comparison with 3.75% indomethacintape, Voltaren_tape was significantly superior in terms of both the AUEC of edema ratio and1/AUEC of pain threshold ratio. Conclusions The results of this study demonstrated that 1% diclofenac sodium(Voltaren_tape)would exert more potent anti−inflammatory and analgesic effects compared to 3.5% or0.5% felbinac containing tapes and 3.75% indomethacin containing tape, using the carrageenan−induced hind footpad paw edema and brewer’s yeast−induced hyper algesia ratmodel. Further clinical investigation is required, but this study suggests that Voltaren_tapemay possibly be a useful formulation for the clinical practice.(Jpn Pharmacol Ther 2009;37:1015−23)KEY WORDS Voltaren_tape, Anti−inflammatory, Analgesic effect, Diclofenac, Felbinac,Indomethacin
