Abstract
Objective To evaluate the effects of capsules containing pentadecyl-containing Aurantiochytrium oil(PAO)on skin conditions in healthy Japanese adults who are concerned about their skin viscoelasticity. Methods In all, 56 subjects were randomly assigned to two groups using a computer program in a 1:1 ratio. They were asked to take one capsule(PAO or placebo)with water without chewing once a day for 6 weeks. One capsule of PAO contained 6.0 mg pentadecyl. The viscoelasticity of the skin, skin surface water content, transepidermal water loss, dermal measurement, visual analog scale of skin conditions, and blood examination were assessed. The primary outcome was the viscoelasticity of the skin(cheeks)at 6 weeks after consumption(6 w). Results The number of subjects ultimately analyzed as full analysis set(FAS)was 28 and 25 for the PAO and placebo groups, respectively. In FAS, the collagen levels significantly increased after the 6-week PAO intervention. In a subgroup analysis of below-average skin surface water content in the FAS, the measured values and amount of change in the skin surface water content were higher with a trend towards significance (group difference:8.7,95%CI (-1.2, 18.5)), and the Intensity of the right cheek were significantly higher in the PAO group than in the placebo group( group difference:811.1,95%CI( 4.3, 17.9)). Conclusions The consumption of one capsule per day containing PAO for 6 weeks improved the collagen levels in the skin of healthy Japanese adults who are concerned about their skin viscoelasticity. The mechanism for the improved skin condition is thought to be the normalization of collagen production due to relaxation of endoplasmic reticulum stress.
